路易·巴斯德(法語:Louis Pasteur,1822年12月27日-1895年9月28日),法國微生物學家、化學家微生物學的奠基人之一以否定自然發生說(自生說)及倡導疾病細菌學說(菌原論)和發明預防接種方法而聞名,為第一個創造狂犬病炭疽疫苗科學家。被世人稱頌為 「進入科學王國的最完美無缺的人」[1]。他和費迪南德·科恩以及羅伯特·科赫一起開創了細菌學,被認為是微生物學的奠基者之一[2],常被稱為「微生物學之父」。[3][4][5]

2005年,法國國家二台舉行了「最偉大的法國人」的評選活動,結果巴斯德名列第二位,僅次於夏爾·戴高樂[6]

生平

巴斯德於1822年生於法國汝拉省的一個小鎮多爾。他父親是拿破崙軍騎兵隊的一名退伍軍人,後成為皮鞋匠,家境並不富裕。巴斯德出生2月後,他們家就搬遷到阿爾布瓦[3]。他在1839年-1842年在貝桑松皇家學院學習,並開始對科學感興趣。由於對科學的興趣,巴斯德從1843年-1846年在巴黎高等師範學校學習,1845年獲碩士學位,1847年獲博士學位畢業。他於1848年開始任第戎大學物理學教授,後在1849年轉至斯特拉斯堡大學理學院任化學教授[7]。在那裡,巴斯德遇到了他的妻子並於1849年5月29日結婚。巴斯德共有過5個子女中,只有兩個活到成年,而其餘三個死於傷寒,此事激發了他去研究治癒各種傳染病的方法。[3][8]他在1868年首次中風,1887年再次中風。1895年9月28日卒於巴黎,享壽72歲。[9]

研究經歷

巴斯德於1854年-1857年任里爾科技大學理學院院長和化學教授,1857年-1867年任巴黎高師的主管行政管理和科學研究的主任,於1862年被選為法國科學院院士,1863年-1867年兼任巴黎美術學校教授,1867年-1874年任巴黎大學化學教授,1869年成為英國皇家學會會員,1867年-1888年任高等師範學校生理化學實驗室主任,1882年被選為法蘭西學院院士,1887年被選為法國科學院永久秘書,1888年-1895年任巴斯德研究所所長。1892年,巴黎大學為慶祝巴斯德70壽辰舉行了盛大的國際性慶典。[7]

研究貢獻

分子不對稱性

早期的巴斯德是一名化學家,還在大學時代,巴斯德便開始了對化學結晶體形態和結構的研究。當時,人們發現從生物合成的酒石酸會令平面偏振光右旋的,但是化學合成得到的酒石酸(異酒石酸)卻沒有旋光性,但是化學分析表明,這兩種途徑得到的酒石酸的化學反應和元素構成都是一模一樣的。通過顯微鏡的觀察,他發現化學合成的異酒石酸里實際上包含了兩種晶體。一種就是常見的右旋的酒石酸,另一種則是左旋的。兩種晶體的混合後,使得旋光性消失。巴斯德在1848年發表了他的發現[10][11]。這一發現證實了旋光異構體的設想,對結構化學的發展具有重要影響。[12][13][14][15][16]巴斯德以後又提出了分子不對稱性理論,開創了立體化學研究的途徑。他還發現生物體對這兩種不對稱性的晶體具有明顯的選擇性。

發酵的細菌理論

法國的葡萄酒業非常有名,但是久置的啤酒和葡萄酒會變酸。里爾釀酒商向巴斯德請教如何防止讓酒變酸。巴斯德首先研究了酒的發酵過程。他發現發酵是因為微生物酵母菌)的增長造成的。酒變酸和發酵類似,不過是由不同的微生物引起的。巴斯德的發現改變了以往認為微生物是發酵的產物,發酵是一個純粹的化學變化過程的錯誤觀點。同時,巴斯德通過大量實驗提出:環境、溫度、pH值和基質的成分等因素的改變,以及有毒物質都以特有的方式影響着不同的微生物。他隨後創立了「巴斯德消毒法」(60~65攝氏度作短時間加熱處理,殺死有害微生物的一種消毒法)並應用在各種食物和飲料上。[7][16]

1862年,巴斯德經多次實驗,終於以鵝頸瓶swan neck duct進行實驗,證明煮沸的肉湯內,不會增長細菌;因此否定認為「生物隨時可由非生物發生」的自然發生說(無生源論、自生論)。他提出「一切生物來自生物」的結論(即生源論)。[17]

免疫學和疫苗

除了發現防止紅酒變酸的方法之外,巴斯德也找到了導致蠶生病的原因。他的研究成果捍衛法國在絲織產業與時尚工業的優勢地位,這又是一樁足以令全法國人向他致敬的傑出成就。他接着將研究範圍拓展到家禽、家畜與人類疾病作用的方式,更透過研究疫苗證實其功效。巴斯德提出了預防接種措施,認為傳染病的微生物在特殊的培養之下可以減輕毒力,變成防病的疫苗。他於1881年著手研究狂犬病,1885年以減毒的方式(the method for attenuatio of virulent microorganisms)研製出減毒狂犬病疫苗,巴斯德的名聲引來大西洋彼岸的求助,當時美國新澤西幾名男童遭到感染狂犬病的犬隻攻擊,性命垂危。這起新聞引起美國民眾的重視,自發集資協助這幾名男童跨越大西洋至巴黎,尋求巴斯德的救助,而巴斯德也不負眾望,利用他研究出的狂犬病疫苗,在同年7月6日治療一受狂犬咬傷的9歲兒童Joseph Meister[7]至此,巴斯德已經是跨越歐陸國界及大西洋隔閡的知識英雄。

信仰和靈性

天主教的觀察者經常說巴斯德一生都是虔誠的基督徒。巴斯德的女婿在他為巴斯德所寫的傳記中也寫過:

他完全相信上帝和永恆,深信今世賦予我們從善的力量將一直持續至來世,這種感情貫穿在巴斯德的整個生活當中。他念念不忘《新約》的福音所闡明的德行教條。就在自己生命的最後幾周里,巴斯德對先人的宗教儀式充滿了敬意。他樸素而自然的以這種儀式作精神支柱。[18]

莫里斯·瓦萊里-拉多Maurice Vallery-Radot是巴斯德女婿兄弟的外孫,也是天主教徒。他也堅持巴斯德深信天主教。[19]有一個被巴斯德·瓦萊里-拉多和莫里斯·瓦萊里-拉多證明是假的所謂巴斯德的話:[20]"The more I know, the more nearly is my faith that of the Breton peasant. Could I but know all I would have the faith of a Breton peasant's wife".[3]根據莫里斯·瓦萊里-拉多,[21]這個錯誤的傳言最早是在巴斯德時候不久出現的。[22]

但巴斯德的外孫路易斯·巴斯德·瓦萊里-拉多Louis Pasteur Vallery-Radot說,巴斯德僅僅相信天主教中的靈性,但沒有參加過日常的宗教活動,[23]儘管巴斯德信仰上帝,一般說,他的觀點更接近自由思想家而不是天主教,他表現更多地是靈性的而不是宗教的。[24][25][26]巴斯德也反對將科學和宗教混在一起。[27][28]

對巴斯德的指控

1995年,在巴斯德逝世百年紀念之際,美國普林斯頓大學科學史教授拉爾德·蓋森Gerald L. Geison出版了《巴斯德的隱秘科學》(The Private Science of Louis Pasteur)一書。書中聲稱,巴斯德這位「最完美無缺的科學家」有欺騙行為,巴斯德的實驗記錄有的不符合他公開的說法,並且他還從事了違反醫學倫理的試驗。[29]紐約時報》立刻報道了該書的出版。[30][31]1962年諾貝爾化學獎得主佩魯茨在《紐約書評》上發布文章《為先驅辯護》進行詳細的反駁。[32]此後,相關爭論一直不斷。[33][34][35]

巴斯德研究所

1887年,巴斯德在巴黎以募捐方式創立了一所公益型私人研究所——巴斯德研究所。建立之初,研究所規模很小,第一批研究隊伍僅設5個實驗室,有5位室主任和14位助手。至2005年,巴斯德研究所擁有一所醫院。該所既是傳染性疾病、熱帶病理學和免疫系統疾病的專業保健中心,又是研究生教育(科研和臨床培訓)中心,生物學基礎研究中心、公共健康應用研究中心。同時它還建有一個博物館、一個包括圖書館在內的科技信息中心、多家公司和分布在全球29個國家內的32所研究機構。[36][37] 共有8位諾貝爾生理學和醫學獎獲得者在該所學習和工作過,他們是拉韋朗梅契尼科夫博爾代尼科爾博韋利沃夫雅各布莫諾[37]

Osler, who knew Pasteur personally. Vallery-Radot was himself a Catholic yet admits that Pasteur believed only in "an Infinite" and "hoped" for a future life. Pasteur publicly stated this himself in his Academy speech in 1822 (in V.R.). He said: "The idea of God is a form of the idea of the Infinite whether it is called Brahma, Allah, Jehova, or Jesus." The biographer says that in his last days he turned to the Church but the only "evidence" he gives is that he liked to read the life of St. Vincent de Paul, and he admits that he did not receive the sacraments at death. Relatives put rosary beads in his hands, and the Catholic Encyclopedia claims him as a Catholic in virtue of the fact and of an anonymous and inconclusive statement about him. Wheeler says in his Dictionary of Freethinkers that in his prime Pasteur was Vice-President of the British Secular (Atheist) Union; and Wheeler was the chief Secularist writer of the time. The evidence is overwhelming. Yet the Catholic scientist Sir Bertram Windle assures his readers that "no person who knows anything about him can doubt the sincerity of his attachment to the Catholic Church," and all Catholic writers use much the same scandalous language.

Pasteur had great respect for the unknown and the infinite, but did not allow himself to become a victim of superstition and fanatical religious explanations.
